Duloxetine is a selective 5-HT and NE reuptake inhibitor (SSNRI). The exact mechanism of the antidepressant and central analgesic effects of duloxetine has not been clarified, but it is believed to be related to the enhancement of serotonin and noradrenergic functions in the central nervous system. Preclinical results show that duloxetine is a strong inhibitor of neuronal 5-HChemicalbookT and NE reuptake. The inhibitory effect on dopamine reuptake is relatively weak. The results of in vitro studies show that duloxetine has no significant affinity with dopaminergic receptors, adrenergic receptors, cholinergic receptors, histaminergic receptors, opioid receptors, glutamate receptors, and GABA receptors. Duloxetine does not inhibit monoamine oxidase.
